产品概述

NGF最初是以7S复合体的形式从小鼠下颌下腺分离得到，该复合体由共价相连的三个亚基α、β和γ组成。其中，α 和γ亚基均是激肽释放酶基因家族（丝氨酸蛋白酶）中的成员；β亚基，也称为β-NGF或2.5S NGF，具备NGF所有的生物学功能。人的重组β-NGF是由两个120个氨基酸的多肽构成的同型二聚体。在氨基酸水平，人的β-NGF蛋白与小鼠、大鼠均有近90%的同源性，并且具有跨物种功能。 NGF是一个典型的嗜神经性蛋白，在外周神经系统中交感神经和部分感觉神经元的发育中发挥重要作用。此外，在中枢神经系统，NGF可作为基底前脑胆碱能神经元的营养因子。NGF对于非神经元组织也具有一定生物学功效。

产品原理

本实验采用双抗体夹心ELISA法。抗人β-NGF单抗包被于酶标板上，标本和标准品中的β-NGF会与单抗结合形成免疫复合物，游离的成分被洗去；加入生物素化的抗人β-NGF抗体，生物素化抗人β-NGF抗体与酶标板上结合的标准品或样本中的β-NGF结合而形成免疫复合物，游离的成分被洗去；加入辣根过氧化物酶标记的亲合素，亲合素与生物素特异性结合，游离的成分被洗去。加入显色底物（显色剂），若反应孔中有β-NGF，辣根过氧化物酶会使无色的显色剂变蓝，加终止液变黄。在450nm处测OD值， β-NGF浓度与OD₄₅₀值之间呈正相关，可通过绘制标准曲线求出标本中β-NGF浓度。

产品性能

1. 灵敏度：最小可测人β-NGF达4pg/ml 。

2. 重复性：板内、板间变异系数均<10%。

产品组分及储存条件

试验所需自备仪器和试剂:

1. 酶标仪(450nm检测波长滤光片，570nm或630nm校正波长滤光片)。

2. 高精度加液器及一次性吸头：0.5-10μl，2-20μl，20-200μl，200-1000μl。

3. 37℃恒温箱、蒸馏水或去离子水、坐标纸。
