肝细胞生长因子（Hepatocyte growth factor，HGF）是一种多效应生长因子，又称扩散因子（scatter factor，SF）、肝细胞生成素A（hepatopoietin A）（肝细胞生成素B可能是A的片段）和乳腺生长因子，最初从大鼠的血小板中纯化获得。成熟的肝细胞生长因子是一个分子量82kDa，含674个氨基酸残基的异二聚体糖蛋白，它是一类因子小家族的一员，该家族还包括肝细胞生长因子样蛋白（也叫巨噬细胞刺激蛋白，MSP），这类因子与其他已知的生长因子几乎没有明显的同源性。肝细胞生长因子对肝细胞和其他上皮细胞，以及内皮细胞、间充质细胞和造血细胞类具有多种不同的作用，包括促进有丝分裂，形态形成和促使细胞运动等。
产品原理
本实验采用双抗体夹心ELISA法。抗人HGF单抗包被于酶标板上，标本和标准品中的HGF会与单抗结合形成免疫复合物，游离的成分被洗去；加入生物素化的抗人HGF抗体，生物素化抗人HGF抗体与酶标板上结合的标准品或样本中的HGF结合而形成免疫复合物，游离的成分被洗去；加入辣根过氧化物酶标记的亲合素，亲合素与生物素特异性结合，游离的成分被洗去。加入显色底物（显色剂），若反应孔中有HGF，辣根过氧化物酶会使无色的显色剂变蓝，加终止液变黄。在450nm处测OD值，HGF浓度与OD450值之间呈正相关，可通过绘制标准曲线求出标本中HGF浓度。
